Both common late-onset familial and sporadic forms of Alzheimer's disease are genetically associated with the APOE gene on chromosome 19, which is responsible for coding apolipoprotein E. -ε2, -ε3, and -ε4 are the three alleles of the human APOE gene, which are distributed in the population at 7%, 79%, and 14%, respectively.



The presence of one or two copies of the ε4 allele in the APOE gene is statistically associated with an elevated risk of developing Alzheimer's disease. Approximately 65% of Alzheimer's disease patients in the Caucasian population possess the APOE ε4 allele.
